ABSTRACT

Background@#Spinal nerve ligation (SNL) model is one of the representative models of the neuropathic pain model. Neuropathic pain in a chronic post-ischemic pain (CPIP) mimics the symptoms of complex regional pain syndrome (CRPS). The administration of polydeoxyribonucleotide (PDRN), which has regenerative and anti-inflammatory effects, has been studied and is used in clinical practice treating various diseases. However, the analgesic effect of PDRN in a neuropathic pain or CRPS model remains unknown. @*Methods@#PDRN (3.3, 10, and 20 mg/kg) was administered into the subcutaneous (SC) layer of the hind paws of SNL and CPIP models. Mechanical anti-allodynic effects were then investigated using the von Frey test. In the immunohistochemical examination, dorsal root ganglia (DRG) and the spinal cord were harvested and examined for the expression of glial fibrillary acidic protein (GFAP) after the 20 mg PDRN injection. @*Results@#Mechanical allodynia was significantly alleviated by administration of PDRN in SNL and CPIP mice at all of the time point. As the dose of PDRN increased, the effect was greater. The 20 mg PDRN injection was found to have the most effective anti-allodynic effect. The increased expression of GFAP in DRG and the spinal cord of SNL and CPIP model decreased following the administration of PDRN than vehicle. @*Conclusion@#SC administration of PDRN results in the attenuation of allodynia and activation of astrocytes in neuropathic pain or CRPS models. We propose that PDRN can have significant potential advantages in neuropathic pain treatment.

ABSTRACT

OBJECTIVE: To determine the validity and reliability of the Korean version of the Coma Recovery Scale-Revised (K-CRSR) for evaluation of patients with a severe brain lesion. METHODS: With permission from Giacino, the developer of the Coma Recovery Scale Revised (CRSR), the scale was translated into Korean and back-translated into English by a Korean physiatrist highly proficient in English, and then verified by the original developer. Adult patients with a severe brain lesion following traumatic brain injury, stroke, or hypoxic brain injury were examined. To assess the inter-rater reliability, all patients were tested with K-CRSR by two physiatrists individually. To determine intra-rater reliability, the same test was re-administered by the same physiatrists after three days. RESULTS: Inter-rater reliability (k=0.929, p < 0.01) and intra-rater reliability (k=0.938, p < 0.01) were both high for total K-CRSR scores. Inter- and intra-rater agreement rates were very high (94.9% and 97.4%, respectively). The total K-CRSR score was significantly correlated with K-GCS (r=0.894, p < 0.01), demonstrating sufficient concurrent validity. CONCLUSION: K-CRSR is a reliable and valid instrument for the assessment of patients with brain injury by trained physiatrists. This scale is useful in differentiating patients in minimally conscious state from those in vegetative state.

ABSTRACT

PURPOSE: The purpose of this study was to examine the effects of parental stress, social support, and coping behavior on burnout among mothers caring for children with cerebral palsy. METHODS: Participants in this cross-sectional, descriptive study were 185 mothers who completed a self-report structured questionnaire. The data were analyzed using descriptive statistics, t-test, one-way ANOVA, correlation and multiple linear regression analyses with IBM SPSS Win 22 program. RESULTS: Parental stress and coping behavior were strong predictors of burnout among mothers of children with cerebral palsy. These variables explained 50.0% of the variance in burnout. Social support was not a predictor of burnout. A higher level of burnout was associated with higher levels of parental stress and lower levels of social support and coping behavior. CONCLUSION: Mothers of children with cerebral palsy are vulnerable to burnout. These results show that effective strategies for reducing parental stress and improving positive coping behavior are needed to reduce burnout in these mothers.

ABSTRACT

OBJECTIVES: We used a questionnaire to measure the oral health literacy of Korean adults. We then evaluated the validity and reliability of the questionnaire. METHODS: This questionnaire was developed using a literacy assessment tool from the National Institute of Korean Language, Republic of Korea. The survey was composed of instructions for dental treatment and oral health information. The components were sub-classified into three cognitive processes (realistic, deductive, and critical domains). Of the 337 university students who received the questionnaire, 84.3 percent responded. We calculated the reliability of the questionnaire using Cronbach's α, to assess the item discrimination and difficulty as well as effectiveness of distracters. RESULTS: We found that the reliability of the 24 items in the questionnaire was 0.89. The mean item discrimination and difficulty values were 0.53 and 0.71. The percentage of correct answers was higher than the effectiveness of the distracters in all items. The mean number of answers to the oral health information data (72.1) was higher than that to dental treatment instructions (63.9). The mean number of answers to the items in the realistic domain (76.1) was higher than that to the items in the deductive (69.4) and critical (65.9) domains. Neither gender nor university major had a significant effect on the percentage of correct answers. CONCLUSIONS: Sixty percent of university students were found to have the government-recommended level of oral literacy. Therefore, better oral health education is required to ensure that all individuals meet government-recommended levels.

Objectives To evaluate possible lipid catabolism and body fat regulation effects of 3-caffeoylquinic acid in Green coffee bean extract (GCBE) in high-fat diet (HFD)-induced obese mice. Methods Obesity was induced in mice using a HFD for four weeks. Then, mice were fed only HFD or HFD with GCBE at 50, 100, and 200 mg/kg. Fatty acid synthesis mechanism regulation of body fat was investigated through real-time PCR and Western blot assay. Body fat reduction was measured through dual-energy X-ray absorptiometry. Results In HFD-induced obese mice, GCBE treatment significantly decreased body weight gain, liver weight and white adipose tissue weights with regulation of adipose tissue lipolysis hormones, like adiponectin and leptin. GCBE treatment decreased mRNA expression levels of adipogenesis and adipocyte metabolism related genes in adipose tissues and the liver, and decreased the corresponding protein expression. Dual energy X-ray absorptiometry measurements were used to compare body fat between mice on high-fat and those treated with GCBE. GCBE treated mice had a lower fat mass compared to HFD alone fed mice and relative body weight and fat mass were markedly decreased. Conclusions GCBE has a potential anti-obesity effect with lowering body fat accumulation by regulating adipogenesis and lipid metabolism-related genes and proteins in WAT and liver.

ABSTRACT

BACKGROUND: The acidic pH of the stratum corneum (SC) is important for epidermal permeability barrier homeostasis. Acidification of the skin surface has been suggested as a therapeutic strategy for skin disorders such as atopic dermatitis (AD). OBJECTIVE: We performed an animal study to evaluate the usefulness of acidification of SC for inhibition of AD lesions and to find out if the therapeutic effect of vinegar is attributable to its herbal contents, rather than its acidity. METHODS: Five groups of six oxazolone-treated (Ox)-AD mice were treated for three weeks with creams of different acidity: vehicle cream alone (pH 5.5), neutralized vinegar cream (pH 7.4), pH 5.0 vinegar cream, pH 3.5 vinegar cream, and pH 3.5 hydrogen chloride (HCl) cream. Also, we have compared two groups of Ox-AD mice treated with pH 5.5 vehicle cream or pH 5.5 vinegar cream. RESULTS: Ox-AD mice treated with acidic creams exhibited fewer AD-like lesions, had significantly lower eczema scores, decreased basal by transepidermal water loss (TEWL), and increased SC hydration compared to the groups given only vehicle and neutral cream. There was no significant difference between the acidic vinegar and HCl groups. Between the groups treated with vehicle and pH 5.5 vinegar cream, there was no difference in eczema score, basal TEWL and SC hydration. CONCLUSION: Application of topical acids, regardless of their source materials, inhibits the development of AD lesions by maintenance of skin surface pH and skin barrier function in murine model.

ABSTRACT

OBJECTIVES: The purpose of this study was to investigate oral discomfort factors in the elderly and to analyze the sociodemographic and health-related characteristics influencing their oral discomfort using multiple regression analysis. METHODS: Facilities were selected based on the 2014 Elderly Welfare Facilities Status. Data was collected through the interview of 176 participants aged ≥65 years. The oral discomfort factors (10 items) were investigated using a 5-point Likert scale. The total oral discomfort scores were added for each of the 10 items investigated using the 5-point Likert scale. SPSS 12.0 program was used to analyze the descriptive statistics and perform a t-test and a stepwise multiple regression analysis. RESULTS: The oral discomfort factor with the highest score was g oral discomfort factorThe total oral discomfort score for women was significantly higher than that for men. 'men. gnificantly higher thangenderg were, respectively, the health-related and sociodemographic characteristics that most significantly influenced the total oral discomfort scores. Perceived oral health accounted for 30.4% of the total oral discomfort score, while gender accounted for 2%. CONCLUSIONS: Health-related characteristics were more significantly associated with oral discomfort than sociodemographic characteristics. Since sociodemographic characteristics are not easily changed, government and healthcare providers should strive to improve the health of the elderly population.

ABSTRACT

Objective: To investigate the anti-obesity activity and the action mechanism of the roots of Adenophora triphylla var. japonica extract (ATE) in high-fat diet (HFD)-induced obese mice and 3T3-L1 adipocytes. Methods: The roots of Adenophora triphylla were extracted with 70% ethanol. To demonstrate the compounds, linoleic acid was analyzed by using gas chromatography; and the anti-obesity effects and possible mechanisms of ATE were examined in 3T3-L1 adipocytes and HFD-induced obese mice. Results: Treatment with ATE inhibited the lipid accumulation without cytotoxicity in 3T3-L1 adipocytes. Furthermore, 200 and 400 mg/kg ATE treatment significantly decreased the body weight gain, white adipose tissues (WATs) weight and plasma triglyceride level, while 100 and 200 mg/kg ATE treatment increased the plasma high-density lipoprotein cholesterol level in the HFD-induced obese mice, as compared with the HFD group. Treatment with 200 and 400 mg/kg ATE also lowered the size of adipocytes in adipose tissue and reduced the lipid accumulation in liver. ATE treatment showed significantly lower expression level of adipogenesis-related proteins, such as peroxisome proliferator-activated receptor γ, fatty acid binding protein (aP2), fatty acid synthase in 3T3-L1 adipocytes; and furthermore, decreased peroxisome proliferator-activated receptor γ, aP2, fatty acid synthase, sterol regulatory element binding protein-1c, and lipoprotein lipase mRNA expression levels in WAT of the HFD-induced obese mice. Conclusions: These results suggested that the ATE has an anti-obesity effect, which may be elicited by regulating the expression of adipogenesis and lipogenesis-related genes and proteins in adipocytes and WAT of the HFD-induced obese mice.

BACKGROUND: The external jugular vein (EJV) is a useful intravenous (IV) cannulation site for anesthesiologists, although it has a relatively high failure rate. Unlike other central veins, visualization of the EJV is important during IV cannulation, and the Valsalva maneuver distends the jugular venous system. However, the relationship between the maneuver and EJV visibility remains unknown. This study compared EJV visibility before and after the Valsalva maneuver. METHODS: This was a prospective observational study that included 200 participants. After the induction of anesthesia, EJV visibility grade, depth from the skin to the EJV superficial surface (EJV depth), and EJV cross-sectional area (CSA) before the Valsalva maneuver were measured. The same parameters were measured after the Valsalva maneuver was performed. The EJV visibility grade was defined as grade A: good appearance and good palpation, grade B: poor appearance and good palpation, and grade C: poor appearance and poor palpation. RESULTS: Patient body mass index and EJV depth affected the EJV visibility grade before the Valsalva maneuver (p < 0.05), although EJV CSA did not. The Valsalva maneuver distended EJV CSA and reduced EJV depth, although these changes were not correlated with EJV visibility grade. With regard to EJV visibility, 34.0% of grade B cases and 20.0% of grade C cases were improved by the Valsalva maneuver. CONCLUSIONS: Although the Valsalva maneuver improved EJV CSA and EJV depth, it did not greatly affect EJV visibility grade.

ABSTRACT

OBJECTIVES: The study on dental fear has increased recently. Studies found that people with high dental fear and or anxiety were less likely to visit dentists. The aim of this study is to review the trend of study on dental fear and to analyse the major factors related to dental fear. METHODS: Seventy four papers listed in Korea Citation Index and SCI(E) were assessed. Quality Assessment and Validity Tool for Correlation Studies (QAVTCS) and Dental Fear Study (DFS) were used in this study. SPSS 12.0 was employed to analyse the descriptive statistics, t-test and Multiple Linear Regression (MLR) with stepwise. RESULTS: The total mean score of QAVTCS for international studies was significantly higher than domestic studies (P=.001). The mean scores of item, 'Seeing the anesthetic needle' and 'Feeling the anesthetic needle', were severe level (Means= 3.05, 3.14, severe>3.00). According to the MLR, the powerful predisposing factors (independent variable) on 'Overall fear of dentistry (dependent variable)' were 'Hearing the drill' (P<.01) and 'Having teeth cleaned' (P<.05). CONCLUSIONS: It was concluded that people had high dental fear in direct dental treatment environment such as contact with seeing anesthetic needle and hearing dental drill. Therefore, this study suggests the need to map out specific and multidirectional plan for decreasing dental fear.

ABSTRACT

OBJECTIVES: Oral health literacy is defined as 'the degree to which individuals have the capacity to obtain, process, and understand basic oral health information and services needed to make an appropriate health decision'. This goal of this study was to develop an instrument for assessing oral health literacy in Korean adults. METHODS: This instrument was developed by using the adult literacy evaluation tool of the Korean Government. The components were mainly related to 2 conditions ('oral health information' and 'dental treatment instruction'), and they were classified into 3 cognitive processes (realistic, deductive, and critical domains). Furthermore, all of the subcomponents were divided into prose and non-prose. Therefore, each of the 12 subcomponents had 4 items, resulting in a final questionnaire with 48 items. A pilot survey was conducted in 51 adults, and the quality of the questionnaire was subsequently evaluated. We identified reliability by using Cronbach's alpha, discrimination, and difficulty. Two items from each of the 12 subcomponents were selected as the final items. We set the difference thresholds at over 0.5 and over 0.3 for reliability and discrimination, respectively. Finally, we distributed the difficulty from 35 to 95%. RESULTS: The reliabilities of all items (48) and the final items (24) were 0.838 and 0.836, respectively. The mean discrimination of the final items was higher than that of all items (mean: 0.5 and 0.3, respectively). The difficulty curve of the final items followed a normal distribution. CONCLUSIONS: The reliability and validity demonstrated by the final 24 items indicates that they are appropriate for evaluating oral health literacy in adults.

METHODS: NEM was administered at 52 mg/kg, 200 mg/kg, and 400 mg/kg to SD-Rat, where arthritis was induced by monosodium iodoacetate (MIA) at 3 mg. NO production in serum was measured using Griess reagent. Cytokines including IL-1beta, and IL-6 were measured by Luminex and PGE2, MMP-2, MMP-9, TIMP-1, LTB4, and hs-CRP were measured by ELISA. The cartilage of patella volume was examined and 3-D high-resolution reconstructions of the cartilage of patella were obtained using a Micro-CT system. RESULTS: Production of NO, IL-1beta, IL-6, PGE2, MMP-2, MMP-9, TIMP-1, LTB4, and hs-CRP in serum was decreased, respectively, in comparison with control. The cartilage of patella volume increased significantly. In addition, the NEM group showed a decrease in the cartilage of patella, synovial membrane, and transformation of fibrous tissue. CONCLUSION: The results for NEM showed significant anti-arthritis activity. These results may be developed as a raw material for new health food to ease the symptoms mentioned above.
